目的建立反相高效液相色潽法(RP-HPLC)同时测定通淋颗粒中绿原酸和咖啡酸的含量。方法采用Kromasil C18色谱柱,以乙腈-0.2%磷酸溶液(11∶89)为流动相,检测波长:323 nm,流速:1 mL·min-1,柱温:40℃,进样量10μL。结果绿原酸和咖啡酸的线性范围分别为1.56~99.60μg·mL-1(r=0.999 5,n=7)和1.68~107.60μg·mL-1(r=0.999 5,n=7);平均加样回收率分别为99.21%~99.80%(RSD均<1%)和99.07%~100.17%(RSD均<2%)。结论所建立的方法简便可靠,专属性强,重现性好,可用于通淋颗粒的质量控制。
Objective To establish an RP-HPLC method for simultaneous determination of chlorogenic acid and caffeic acid in Tonglin Granules. Methods The mobile phase was acetonitrile-0.2% phosphoric acid solution (11:89) with Kromasil C18 column. The detection wavelength was 323 nm, the flow rate was 1 mL · min-1, the column temperature was 40 ℃ and the injection volume was 10 μL. Results The linear ranges of chlorogenic acid and caffeic acid were 1.56 ~ 99.60μg · mL -1 (r = 0.999 5, n = 7) and 1.68 ~ 107.60μg · mL -1 (r = 0.999 5, n = 7) The average recoveries were 99.21% -99.80% (RSD <1%) and 99.07% -100.17% (RSD <2%), respectively. Conclusion The established method is simple, reliable, specific and reproducible. It can be used for the quality control of Tonglin granules.
