目的观察强肌健力方对脾肾两虚证大鼠胸腺组织的作用及机理。方法将72只SD大鼠随机分为5组,即正常对照组(10只),模型组(16只),强肌健力方组(15只),黄芪减量方组(15只),单味黄芪组(16只)。采用灌服大黄复制脾虚模型(14天),在脾虚基础上采用肌肉注射氢化可的松复制肾虚模型(10天)。造模同时分别给予蒸馏水、强肌健力方、黄芪减量方、单味黄芪灌胃,连续给药24d后,取胸腺组织行HE染色,光学显微镜下观察各组大鼠胸腺组织形态学的变化,用免疫组化的方法测定增殖细胞核抗原(PCNA)表达。结果模型组体质量比正常对照组显著减轻(P<0.01),而各用药组均可使大鼠体质量升高(P<0.05或P<0.01);模型组大鼠胸腺组织有不同程度损伤,而强肌健力方组、单味黄芪组能使已遭损伤的胸腺组织得以修复;与正常对照组比较,模型组胸腺组织PCNA蛋白表达减少,阳性面积率显著降低(P<0.01);而强肌健力方能使胸腺组织PCNA表达升高(P<0.05)。结论强肌健力方能有效促进脾肾两虚证大鼠胸腺组织细胞增殖,并对受损的胸腺组织具有保护作用,其健脾益气的作用机理与升高PCNA表达有关。
Objective To observe the effect and mechanism of Qiangji Jianli Fang on thymus tissue of rats with deficiency of spleen and kidney. Methods Seventy-two SD rats were randomly divided into five groups: normal control group (10 rats), model group (16 rats), Qiangji Jianlifang group (15 rats), and Huangqi decoction group (15 rats). Single-flavored Astragalus group (16). Rhubarb was used to irrigate spleen deficiency model (14 days) and intramuscular injection of hydrocortisone was used to replicate the kidney deficiency model (10 days) on the basis of spleen deficiency. At the same time, the rats were given distilled water, strong muscle Jianli Fang, Huangqi Xiaofang Fang, and single-flavored jaundice. After 24 days continuous administration, the thymus tissues were stained with HE and the thymus histomorphology was observed under light microscope. Changes were detected by immunohistochemical method for proliferating cell nuclear antigen (PCNA) expression. Results The body weight of the model group was significantly lower than that of the normal control group (P<0.01), and the body weight of the rats in each group increased (P<0.05 or P<0.01). The thymus tissue of the model group was damaged to varying degrees. However, the strong muscle-strengthening group and single-flavored astragalus group could repair the damaged thymus tissue. Compared with the normal control group, the expression of PCNA protein in the thymus tissue of the model group decreased, and the positive area ratio decreased significantly (P<0.01). However, strong muscle strength can increase the expression of PCNA in thymus tissue (P<0.05). Conclusion Qiangqiangjianli can effectively promote the proliferation of thymus tissue in spleen and kidney deficiency rats and protect the damaged thymus tissue. The mechanism of strengthening spleen Qi is related to increasing the expression of PCNA.
