目的:采用反相高效液相色谱(HPLC)技术,探讨中药附子对正常大鼠肝组织中三磷酸腺苷(ATP),二磷酸腺苷(ADP)及一磷酸腺苷(AMP)含量及其对能荷(EC)水平的影响。方法:将20只Wister大鼠随机分为2组,分别灌胃附子水煎液和生理盐水4周,4周后麻醉处死,取肝组织进行反相高效液相色谱测定。结果:附子给药组大鼠较对照组大鼠肝脏中ATP含量明显升高,提高了70.3%,具有极显著性差异(P<0.01),ADP,AMP的含量2组间无明显变化,总腺苷酸库、EC水平附子给药组与对照组比亦显著性升高(P<0.05)。结论:热性中药附子可以增加机体能量储备,能显著提高机体肝组织的ATP含量和EC水平,起到影响肝脏能量代谢的作用。
Objective: To study the effect of aconite on the content of adenosine triphosphate (ATP), adenosine diphosphate (ADP) and adenosine monophosphate (AMP) in liver of normal rats and its effect on the energy charge by reversed-phase high performance liquid chromatography (EC) level. Methods: Twenty Wister rats were randomly divided into 2 groups. The rats were administered with aconite decoction and normal saline for 4 weeks respectively. After 4 weeks, the rats were killed by anesthesia and the liver tissue was taken for reverse phase high performance liquid chromatography. Results: The content of ATP in the aconite administration group was significantly higher than that in the control group (70.3%) (P <0.01), while there was no significant difference in the content of ADP and AMP between the two groups Adenosine pool, EC level of aconite administration group and the control group also significantly increased (P <0.05). CONCLUSION: The aconite of heat can increase the body’s energy reserve, significantly increase the level of ATP and EC in the liver of the body, and play an important role in affecting the liver’s energy metabolism.
