采集符合标准的心脏150例,分为3组,实验组为不明原因心源性猝死(SCD)组50例,阴性对照组(交通事故和高坠死亡)50例,阳性对照组(冠心病)50例。应用免疫组化及Simple PCI图像分析系统,比较不明原因SCD组与阴性对照组,以及冠心病组与阴性对照组窦房结和房室结中Cx43、Cx45的表达,并用SPSS 22.0统计软件对数据进行统计分析。房室结和窦房结中Cx43、Cx45的表达:冠心病组与阴性对照组比较,表达数量减少;不明原因SCD组与阴性对照组比较,表达数量减少,通过统计学分析比较,P<0.05,均有统计学意义。本研究实验提示Cx43、Cx45表达数量的减少可能引起细胞间电生理功能障碍,诱发心律失常导致猝死,Cx43、Cx45的免疫组化结果可以成为辅助诊断不明原因SCD的参考指标。
Fifty cases of SCD group, 50 cases of negative control group (traffic accident and high crash death) and 50 cases of positive control group (CHD) were enrolled in this study. 50 cases. Immunohistochemistry and Simple PCI image analysis system were used to compare the expression of Cx43 and Cx45 in sinoatrial node and atrioventricular node between SCD group and negative control group and coronary heart disease group and negative control group. SPSS 22.0 software was used to analyze the data conduct statistical analysis. The expression of Cx43 and Cx45 in atrioventricular node and sinoatrial node: Compared with negative control group, the expression of Cx43 and Cx45 in coronary heart disease group was decreased; the number of expression was decreased in unexplained SCD group and negative control group, P <0.05 by statistical analysis , All with statistical significance. Our study suggests that the reduction of Cx43 and Cx45 expression may lead to electrophysiological dysfunction among cells and sudden cardiac death caused by arrhythmia. The immunohistochemical results of Cx43 and Cx45 may be used as a reference index for diagnosis of unexplained SCD.
