目的:银丹心脑临床治疗冠心病心绞痛疗效显著,但其机理尚不明确。本实验通过观察银丹心脑通对巨噬细胞分泌基质金属蛋白酶的作用,探讨银丹心脑通的作用机理。方法:生长状态良好的单核细胞分化成巨噬细胞,加入不同浓度的药物。24 h后收集细胞上清,ELISA检测细胞上清中MMP-2、MMP-9的含量。结果:给药组不同剂量均能抑制MMP-2、MMP-9的表达(P<0.01-0.05),其中高剂量组效果最为显著。结论:银丹心脑通能够抑制巨噬细胞基质金属蛋白酶的分泌,具有明显的抗炎作用,此实验为其在临床上的进一步应用提供了依据。
Objective: Yin Dan heart and brain clinical treatment of coronary heart disease angina significant effect, but its mechanism is not yet clear. In this experiment, we observed the effect of Yindan Xinnaotong on the secretion of matrix metalloproteinases by macrophages and explored the mechanism of Yindan Xinnaotong. Methods: Monocytes with good growth differentiated into macrophages, adding different concentrations of drugs. After 24 h, the supernatant of the cells was collected. The contents of MMP-2 and MMP-9 in the supernatant of the cells were detected by ELISA. Results: The different doses of the drug group could inhibit the expression of MMP-2, MMP-9 (P <0.01-0.05), of which the high-dose group was the most effective. Conclusion: Yin Dan Xin Nao Tong can inhibit the secretion of matrix metalloproteinase in macrophages, and has obvious anti-inflammatory effect. This experiment provides a basis for its further clinical application.
