目的探讨芹菜素对游离脂肪酸(free fatty acids,FFA)诱导的L02细胞的非酒精性脂肪性肝炎(nonalcoholic steatohepatitis,NASH)模型的炎症反应和胰岛素抵抗的治疗活性。方法 FFA处理L02细胞24 h,建立NASH细胞模型;MTT法检测细胞生长抑制率并制定安全给药浓度;油红O染色法观察细胞脂质沉积情况;Elisa法检测细胞的白介素-8(IL-8)、肿瘤坏死因子-α(TNF-α)和葡萄糖转运蛋白-4(GLUT-4)含量;Western-blot法及Elisa法检测细胞的胰岛素受体(IR)的含量。结果 MTT试验显示芹菜素最高给药浓度(280.00μg·m L?1)下,细胞生长抑制率仅为18.8%;与模型组比较,芹菜素高、中、低剂量组(250,150,100μg·m L?1)GLUT-4和IR的含量相均明显提高(P<0.01),而高、中剂量组的IL-8、TNF-α含量和脂变率均明显下降(P<0.01)。结论芹菜素可调节细胞脂变率和IL-8、TNF-α、GLUT-4、IR的含量,改善FFA诱导的L02细胞NASH模型的胰岛素抵抗及炎症反应,且具有极小的细胞毒性。
Objective To investigate the therapeutic activity of apigenin against inflammation and insulin resistance in nonalcoholic steatohepatitis (NASH) model induced by free fatty acids (FFA) in L02 cells. Methods L02 cells were treated with FFA for 24 h to establish NASH cell model. The cell growth inhibition rate was determined by MTT assay and safe administration concentration was established. Lipid deposition was observed by oil red O staining. The levels of interleukin-8 (IL- 8, TNF-α and GLUT-4 were detected by ELISA. The content of insulin receptor (IR) in cells was detected by Western-blot and Elisa method. Results MTT assay showed that the cell growth inhibition rate was only 18.8% under the highest concentration of apigenin (280.00μg · m L · 1). Compared with the model group, the inhibitory rates of apigenin at high, middle and low doses (250,150,100μg · m L 1) The contents of GLUT-4 and IR were significantly increased (P <0.01), while the contents of IL-8, TNF-α and lipid in high and middle dose groups were significantly decreased (P <0.01). Conclusions Apigenin can regulate the lipid peroxidation and the levels of IL-8, TNF-α, GLUT-4 and IR, and improve the insulin resistance and inflammatory response induced by FFA in NASH model of L02 cells with minimal cytotoxicity.
