目的 通过观察脑源性神经生长因子 (BDNF)对顺铂诱导的神经母细胞瘤 (NB)细胞株生长与凋亡的影响 ,探讨TrKB BDNF通路在NB化疗耐药中的作用。方法 应用四甲基偶氮蓝比色 (MTT)法检测不同浓度BDNF对顺铂诱导人NB细胞株SH SY5Y的生长的影响 ,应用流式细胞仪 (FCM )检测BDNF对顺铂诱导的人NB细胞株SH SY5Y的凋亡率的影响。结果 BDNF可阻断顺铂对TrkB表达的SH SY5Y细胞的杀伤作用 ,且其阻断能力的大小与BDNF浓度有关 ,但不能阻断顺铂对无TrkB表达的SH SY5Y细胞的杀伤作用。顺铂对NB细胞的杀伤作用是通过诱导细胞凋亡 ,BDNF对顺铂这一作用的阻断是通过抑制细胞凋亡而实现的。结论 TrkB BDNF通路在TrkB表达的NB化疗耐药机制中起重要作用。
Objective To observe the effects of BDNF on the growth and apoptosis of cisplatin-induced neuroblastoma (NB) cell lines and to explore the role of TrKB BDNF pathway in the chemoresistance of NB chemotherapeutics. Methods The effects of different concentrations of BDNF on the growth of SH-SY5Y cells induced by cisplatin were detected by MTT assay. Flow cytometry (FCM) was used to detect the effect of BDNF on cisplatin-induced human NB Cell line SH SY5Y apoptosis rate. Results BDNF could block cisplatin-induced cytotoxicity of TrkB-expressing SH SY5Y cells. The blocking effect of BDNF was related to the concentration of BDNF, but it could not inhibit the killing effect of cisplatin on SH SY5Y cells without TrkB expression. The killing effect of cisplatin on NB cells is through the induction of apoptosis, and the blockade of the effect of BDNF on cisplatin is achieved by inhibiting apoptosis. Conclusion The TrkB BDNF pathway plays an important role in the mechanism of NB chemoresistance induced by TrkB.
