目的探讨柴胡皂甙D对人宫颈癌Hela细胞系凋亡的影响及相关机制;方法以不同浓度(0、2.5、5、7.5μg/ml)处理人宫颈癌Hela细胞系,MTT法检测处理24h、48h、72h后细胞抑制率;收集经不同浓度柴胡皂甙D处理48h的细胞,利用流式细胞术检测细胞凋亡率;提取经不同浓度柴胡皂甙D处理48h的细胞总蛋白,west-ern blot检测柴胡皂甙D对人宫颈癌Hela细胞系survivin表达的影响;结果柴胡皂甙D对人宫颈癌Hela细胞系细胞活力具有明显抑制作用,并呈浓度和时间依赖方式;经柴胡皂甙D处理48h后,随药物浓度增加,细胞凋亡率也逐渐升高;Western blot表明柴胡皂甙D可抑制人宫颈癌Hela细胞系survivin的表达。结论柴胡皂甙D可诱导人宫颈癌Hela细胞系发生凋亡,这种作用可能与其抑制survivin的表达有关。
Objective To investigate the effects of saikosaponin D on apoptosis in human cervical cancer Hela cell lines and its related mechanisms. Methods Human cervical cancer Hela cell lines were treated with different concentrations (0, 2.5, 5, 7.5 μg / ml) , 48h, 72h after cells were treated with different concentrations of saikosaponin D. The apoptosis rate of cells treated with different concentrations of saikosaponin D for 48h was determined by flow cytometry. Total proteins of cells treated with different concentrations of saikosaponin D for 48h were collected, ern blot was used to detect the effect of saikosaponin D on the expression of survivin in human cervical cancer Hela cell line.Results Saikosaponin D significantly inhibited the viability of human cervical cancer Hela cell line in a concentration- and time-dependent manner; saikosaponin D treatment 48h, with the drug concentration increased, the rate of apoptosis also gradually increased; Western blot showed that saikosaponin D can inhibit the expression of human cervical cancer Hela cell line survivin. Conclusion Saikosaponin D can induce apoptosis of human cervical cancer Hela cell line, which may be related to the inhibition of survivin expression.
