目的探讨23-羟基桦木酸(23-HBA)对体外血管生成的抑制作用。方法采用磺酰罗丹明B(SRB)法测定23-HBA对人毛细血管内皮细胞(HMEC)增殖、迁移和小管形成的影响;用免疫组化染色法检测HMEC中CD31表达的变化。结果23-HBA体外可明显抑制HMEC增殖(IC50为40.44mg/L)、迁移和小管的形成,且呈明显的剂量依赖性。23-HBA的浓度为10mg/L时,HMEC中CD31的表达明显降低。结论23-HBA体外具有明显抑制血管生成的作用,有可能成为有效的血管生成抑制剂。
Objective To investigate the inhibitory effect of 23-hydroxy betulinic acid (23-HBA) on angiogenesis in vitro. Methods The effects of 23-HBA on the proliferation, migration and tubular formation of human capillary endothelial cells (HMECs) were determined by sulforhodamine B (SRB). The expression of CD31 in HMECs was detected by immunohistochemical staining. Results 23-HBA significantly inhibited HMEC proliferation (IC50 = 40.44 mg / L), migration and tubule formation in vitro and in a dose-dependent manner. When the concentration of 23-HBA was 10 mg / L, the expression of CD31 in HMEC was significantly reduced. Conclusion 23-HBA has a significant inhibitory effect on angiogenesis in vitro and may be a potent angiogenesis inhibitor.
