目的建立HPLC同时测定丹酚酸B和阿魏酸的方法。方法色谱柱ODS-C18(4.6 mm×250 mm,5μm)柱,流动相:甲醇-乙腈-甲酸-水,检测波长为286 nm,流速0.8 mL.min?1。结果丹酚酸B浓度在5.40~43.25μg.mL?1,阿魏酸浓度在3.37~27.00μg.mL?1内线性关系良好。丹酚酸B和阿魏酸的线性方程分别为:Y=0.971 1X–2.215 4,r=0.999 8,Y=0.415 9X–0.450 7,r=0.999 9;平均回收率分别为98.4%,98.7%;RSD分别为3.39%,1.49%。结论本法在检测丹酚酸B和阿魏酸时有较好的专属性,灵敏度高,重复性好,且样品处理简单、容易操作,可以作为丹参和当归复方制剂的质量检测方法。
Objective To establish a method for simultaneous determination of salvianolic acid B and ferulic acid by HPLC. Methods Column ODS-C18 (4.6 mm × 250 mm, 5 μm) was used. The mobile phase was methanol-acetonitrile-formic acid-water and the detection wavelength was 286 nm and the flow rate was 0.8 mL.min -1. Results The concentration of salvianolic acid B was in the range of 5.40 ~ 43.25μg.mL -1, and the concentration of ferulic acid was good in the range of 3.37 ~ 27.00μg.mL -1. The linear equations of salvianolic acid B and ferulic acid were as follows: Y = 0.971 1X-2.215 4, r = 0.999 8, Y = 0.415 9X-0.450 7, r = 0.999 9, and the average recoveries were 98.4% and 98.7% ; RSD was 3.39% and 1.49% respectively. Conclusion This method has good specificity in the detection of salvianolic acid B and ferulic acid. It has high sensitivity and good repeatability. The sample is simple to handle and easy to operate. It can be used as the quality test method for the compound preparations of Radix Salviae Miltiorrhizae and Angelica sinensis.
