目的:探讨黄芩素(Baicalein)对人乳腺癌MDA-MB-231细胞肺转移的影响及相关的作用机制。方法:建立人乳腺癌MDA-MB-231细胞裸鼠肺转移模型并给与黄芩素药物干预。实验分为四组:1.对照组(给予生理盐水);2.实验组(给予黄芩素100mg·kg-1);3.低剂量预防组(黄芩素50mg·kg-1);4.高剂量预防组(黄芩素100mg·kg-1)。采用免疫组化SP法和Western blot法,检测裸鼠的肺组织中SATB1,Wnt1,β-catenin,E-cadherin,Vimentin和Snail蛋白的表达。结果:免疫组化结果表明黄芩素可以抑制SATB1,Wnt1,β-catenin,Vimentin,Snail蛋白的表达和促进E-cadherin蛋白的表达。Western blot结果表明黄芩素下调SATB1,Wnt1,β-catenin,Vimentin,Snail蛋白的表达和上调E-cadherin蛋白的表达。各组蛋白平均表达水平差异均有统计学意义(P<0.05)。结论:黄芩素(Baicalein)抑制乳腺癌MDA-MB-231细胞的肺转移,可能机制是抑制SATB1蛋白的表达,阻止Wnt1/β-catenin信号通路,抑制EMT的发生,从而减少乳腺癌肺转移的发生。
Objective: To investigate the effects of baicalein on lung metastasis of human breast cancer MDA-MB-231 cells and its related mechanism. Methods: The lung metastasis model of human breast cancer MDA-MB-231 cells was established and the intervention of baicalein was given. The experiment was divided into four groups: 1. Control group (given saline); 2. Experimental group (given baicalein 100mg · kg-1); 3. Low dose preventive group (baicalein 50mg · kg-1) Dose prevention group (baicalein 100mg · kg-1). The expression of SATB1, Wnt1, β-catenin, E-cadherin, Vimentin and Snail in the lung tissue of nude mice was detected by immunohistochemical SP method and Western blot. Results: The results of immunohistochemistry showed that Baicalein could inhibit the expression of SATB1, Wnt1, β-catenin, Vimentin and Snail protein and promote the expression of E-cadherin protein. Western blot results showed that baicalein down-regulated the expressions of E-cadherin and protein of SATB1, Wnt1, β-catenin, Vimentin and Snail. The average expression level of each group of proteins were statistically significant (P <0.05). CONCLUSION: Baicalein inhibits the lung metastasis of breast cancer MDA-MB-231 cells by inhibiting the expression of SATB1 protein, blocking the Wnt1 / β-catenin signaling pathway, inhibiting the development of EMT and decreasing the metastasis of breast cancer occur.
