目的 观察诱导分化剂苯乙酸对胶质瘤细胞 C6的增殖分化过程中癌基因 C- myc,抑癌基因 P1 6的蛋白水平变化。方法 3H- Td R掺入法测细胞增殖率 ;免疫组化 SP法检测 C- myc、P1 6基因蛋白水平的表达。结果 应用苯乙酸后 ,细胞 CPM值降低 ;C- myc基因胞浆阳性表达下降 ,P1 6基因胞浆阳性表达升高。结论 苯乙酸对胶质瘤细胞存在剂量依赖性抑制 ,作用方式为抑制胞浆中 RNA合成。这些作用与苯乙酸降低了 C- myc基因对细胞恶性增殖的转录调节作用 ,增强了抑癌基因 P1 6活性有关。
Objective To observe the changes of protein level of oncogene C-myc and tumor suppressor gene P1 6 induced by phenylacetate in differentiation and proliferation of C6 glioma cells. Methods 3H-Td R incorporation assay was used to measure the cell proliferation rate. The expression of C-myc and P1 6 protein was detected by immunohistochemical SP method. Results After applying phenylacetic acid, the cell CPM value decreased; the expression of cytoplasm of C-myc gene decreased, and the positive expression of P1 6 gene cytoplasm increased. Conclusion Phenylacetate inhibits glioma cells in a dose-dependent manner by inhibiting the synthesis of RNA in cytoplasm. These effects and phenylacetic acid reduce the transcriptional regulation of C-myc gene on malignant proliferation of cells and enhance the activity of tumor suppressor gene P1 6.
