目的:观察阿霉素肾病大鼠肾小球足细胞CD2相关蛋白(CD2-associated protein,CD2AP)和肾病蛋白(nephrin)的表达及鲤鱼汤对其表达的影响,进而探讨鲤鱼汤治疗肾病综合征的作用机制。方法:70只Wistar大鼠随机分为5组:正常对照组(N组)、模型组(M组)、福辛普利组(F组)、鲤鱼汤低剂量组(YD组)和鲤鱼汤高剂量组(YG组)。M组、F组、YD组和YG组大鼠采用尾静脉注射阿霉素6.5mg/kg复制阿霉素肾病模型。第3周开始进行干预,YG组和YD组分别予以高、低剂量鲤鱼汤(2.25、1.13g/mL)灌胃7周;F组予以0.09mg/mL福辛普利灌胃;N组和M组予以等量自来水灌胃。采用免疫组化方法检测各组大鼠肾脏中CD2AP、nephrin的表达,并计算其阳性表达的平均光密度值(AOD)。结果:与模型组相比,鲤鱼汤高剂量和鲤鱼汤低剂量组大鼠血清总蛋白水平、血清白蛋白水平等均有一定升高,尤其鲤鱼汤高剂量组差异均有统计学意义(P<0.05);各治疗组肾小球中CD2AP与nephrin的表达较N组均降低,但与M组比较,都有不同程度的增加(P<0.05)。结论:鲤鱼汤可不同程度地增强阿霉素肾病大鼠肾小球足细胞CD2AP和nephrin的表达,改善阿霉素肾病大鼠肾小球的损伤,进而起到治疗作用。
Objective: To observe the expression of CD2-associated protein (CD2AP) and nephrin in glomerular podocyte of adriamycin-induced nephropathy rats and the effect of carp soup on its expression, then to explore the effect of carp soup in treating nephrotic syndrome The mechanism of action. Methods: Seventy Wistar rats were randomly divided into five groups: normal control group (N group), model group (M group), fosinopril group (F group), carp soup low dose group (YD group) and carp soup High dose group (YG group). M, F, YD and YG groups were injected with doxorubicin (6.5 mg / kg) into the adriamycin nephropathy model. The rats in YG group and YD group were given high or low dose carp soup (2.25,1.13g / mL) for 7 weeks respectively, while those in F group were given 0.09mg / mL fosinopril. Rats in group N and M group to the same amount of tap water gavage. Immunohistochemistry was used to detect the expression of CD2AP and nephrin in the kidneys of each group, and the average optical density (AOD) value was calculated. Results: Compared with the model group, the serum total protein and serum albumin levels of high-dose carp soup and low-dose carp decoction group increased to a certain extent, especially in the high-dose carp soup group (P <0.05). The expression of CD2AP and nephrin in the glomeruli in each treatment group was lower than that in the N group (P <0.05). Conclusion: The carp soup can enhance the expression of CD2AP and nephrin in glomerular podocyte of adriamycin-induced nephropathy rats to varying degrees and improve the glomerular injury of adriamycin-induced nephropathy rats, and then play a therapeutic role.
