目的:观察安胰颗粒对重症胰腺炎大鼠NF-κB的影响及其与细胞因子的关系,探讨其对SAP的治疗作用机制。方法:将120只大鼠分为正常组(N组)、模型组(SAP组)、安胰颗粒组(A组)和IL-10组(I组)共4组,各组又分为3小时、6小时、12小时三个亚组。予腹腔内注射6%的左旋精氨酸(L-Arginine)溶液建立SAP大鼠模型。I组予腹腔注射10000U人重组白介素-10(rhIL-10)预处理后诱导SAP。A组连续3天予安胰颗粒4g/kg灌胃后诱导SAP。观察胰腺组织病理学变化,免疫组化检测胰腺组织NF-κBp65的表达,酶联免疫吸附法(ELISA)检测血清TNF-α、IL-1β水平。结果 (1)A组及I组病理学评分较SAP组明显下降(P<0.05)。(2)SAP组胰腺组织NF-κBp65各时点表达显著高于N组(P<0.01);A组NF-κBp65各时点表达较SAP组明显下降(P<0.05),12小时时点表达明显低于I组(P<0.05)。(3)与SAP组相比,A组和I组TNF-α及IL-1β浓度明显下降(P<0.05或P<0.01)。(4)胰腺组织NF-κB p65的表达量与血清TNF-α、IL-1β浓度呈明显正相关关系(P<0.05或P<0.01)。结论:安胰颗粒通过抑制NF-κB活化,减少细胞因子的产生和释放,进而改善胰腺损伤,减轻SAP的炎症反应,有效治疗SAP。
Objective: To observe the effect of AnYi granule on NF-κB in rats with severe pancreatitis and its relationship with cytokines, and to explore its mechanism of action on SAP. Methods: 120 rats were divided into normal group (N group), model group (SAP group), pancreas group (group A) and IL-10 group (group I) Hours, 6 hours, 12 hours three subgroups. The rat model of SAP was established by intraperitoneal injection of 6% L-arginine solution. Group I was intraperitoneally injected 10000U human recombinant interleukin-10 (rhIL-10) pretreatment induced SAP. A group for 3 consecutive days to the pancreatic juice pancreas 4g / kg gavage after induction of SAP. The pathological changes of pancreas were observed. The expression of NF-κBp65 in pancreatic tissue was detected by immunohistochemistry. The levels of TNF-α and IL-1β in serum were detected by enzyme linked immunosorbent assay (ELISA). Results (1) The pathological score of group A and group I was significantly lower than that of SAP group (P <0.05). (2) The expression of NF-κBp65 in pancreas of SAP group was significantly higher than that in N group at each time point (P <0.01). The expression of NF-κBp65 at each time point in SAP group was significantly lower than that in SAP group (P <0.05) Significantly lower than the I group (P <0.05). (3) Compared with SAP group, the concentrations of TNF-α and IL-1β in group A and group I were significantly decreased (P <0.05 or P <0.01). (4) The expression of NF-κB p65 in pancreatic tissue was positively correlated with the levels of serum TNF-α and IL-1β (P <0.05 or P <0.01). CONCLUSION: AnYi granule can effectively treat SAP by inhibiting the activation of NF-κB, decreasing the production and release of cytokines, thereby improving the pancreatic injury, relieving the inflammatory reaction of SAP.
