目的建立绵茵陈中绿原酸的含量测定方法并采用薄层色谱-生物自显影法研究绵茵陈提取液的抗氧化活性。方法薄层扫描法测定绵茵陈提取液中绿原酸的含量,随后用二苯代苦味肼基自由基溶液显色,双波长扫描,获得其抗氧化成分的峰面积。结果常规薄层扫描与生物自显影扫描测定得到的绿原酸含量具有一致性,由生物自显影扫描法测得色谱峰总积分值明显大于绿原酸单峰面积,表明绵茵陈中除了绿原酸外,还存在其它抗氧化能力较强的活性成分。结论以DPPH作显色剂,双波长扫描,可利用TLC-生物自显影技术对天然提取物中具有自由基清除及抗氧化活性的活性成分进行快速筛选与评价。
OBJECTIVE To establish a method for the determination of chlorogenic acid in Mian Yin Chen and study its anti-oxidative activity using thin-layer chromatography-autobiography. Methods Thin layer scanning method was used to determine the content of chlorogenic acid in the extract of Mian-yin Chen, followed by the color development of the benzodiazepine hydrazide free radical solution and the double-wavelength scanning to obtain the peak area of the antioxidant component. Results The contents of chlorogenic acid obtained by conventional thin-layer scanning and autoradiographic scanning were consistent. The total integral of chromatographic peaks measured by autoradiography was significantly higher than that of chlorogenic acid, indicating that in addition to green In addition to the original acids, there are other active ingredients that are more resistant to oxidation. Conclusion DPPH as a color reagent, dual-wavelength scanning, TLC-autoradiography can be used for natural extracts of natural extracts with free radical scavenging and antioxidant activity of the active ingredients for rapid screening and evaluation.
