目的比较外周血γδT细胞和αβT细胞对胰腺癌细胞株的杀伤作用,研究γδT细胞在胰腺癌过继免疫治疗中的作用。方法从胰腺癌病人外周血中分离出淋巴细胞,利用固相抗体包被方法体外特异性扩增γδT和αβT细胞,用流式细胞仪检测效应细胞的纯度,按一定的靶效比将效应细胞和胰腺癌细胞株体外混合培养。利用 MTT 法测定效应细胞的细胞毒作用。结果利用抗体包被方法可有效扩增γδT和αβT细胞,并达到较高纯度。外周血γδT细胞体外杀伤胰腺癌细胞株的作用与效应细胞纯度和靶效比成正相关,并对多株胰腺癌细胞株敏感;外周血γδT细胞的杀伤效果明显优于αβT细胞。结论外周血γδT细胞可以弥补传统效应细胞的不足,表现出较强的肿瘤免疫杀伤作用。
Objective To compare the killing effect of γδT cells and αβT cells on pancreatic cancer cell lines and to study the role of γδT cells in adoptive immunotherapy of pancreatic cancer. Methods Lymphocytes were isolated from the peripheral blood of patients with pancreatic cancer. The γδT and αβ T cells were specifically amplified by solid phase antibody-coated method. The purity of effector cells was detected by flow cytometry. The effector cells And pancreatic cancer cell lines in vitro mixed culture. The cytotoxic effect of effector cells was measured by MTT assay. Results The antibody-coated method could effectively amplify γδT and αβT cells with high purity. The effect of peripheral blood γδT cells killing pancreatic cancer cell lines in vitro was positively correlated with effector cell purity and target efficiency, and was sensitive to many pancreatic cancer cell lines. The cytotoxicity of γδT cells in peripheral blood was significantly better than that of αβT cells. Conclusion The peripheral blood γδT cells can make up for the lack of traditional effector cells and show a strong tumor immune killing effect.
