目的:探讨冲击波对人骨髓间充质干细胞(MSC)增殖和成骨分化能力的影响。方法:从健康志愿者骨髓中分离培养骨髓间充质干细胞,将其分为3组,空白对照组细胞用普通培养液培养,成骨诱导组细胞用经典成骨诱导剂培养,冲击波组细胞用普通培养液培养,外加冲击波刺激。刺激后在镜下观察MSC形态;应用CCK-8检测冲击波组细胞和空白对照组MSC增殖能力的变化;第14天和28天分别用碱性磷酸酶和Von Kossa染色检测各组细胞的成骨能力。结果:CCK-8检测发现,冲击波与空白对照组相比,细胞的增殖活性显著增强;碱性磷酸酶和Von Kossa染色结果表明,冲击波组细胞的成骨分化能力比成骨诱导组要高;空白对照组细胞染色呈现弱阳性。结论:冲击波能促进骨髓间充质干细胞的增殖能力和成骨分化能力。
Objective: To investigate the impact of shock wave on human bone marrow mesenchymal stem cells (MSC) proliferation and osteogenic differentiation. Methods: Bone marrow-derived mesenchymal stem cells (MSCs) were isolated from bone marrow of healthy volunteers and divided into three groups. The cells in blank control group were cultured in normal medium. The cells in osteogenic group were cultured with classical osteoinductive agent. Common culture medium, plus shock wave stimulation. The morphological changes of MSCs were observed under light microscopy after stimulation. The proliferation of cells in shock wave group and blank control group were detected by CCK-8. Osteoblasts were detected by alkaline phosphatase and Von Kossa staining on day 14 and 28, respectively ability. Results: The CCK-8 assay showed that the proliferation activity of shock wave was significantly higher than that of blank control group. Alkaline phosphatase and Von Kossa staining showed that the osteogenic differentiation ability of shock wave group was higher than that of osteogenic induction group. Blank control cells showed weakly positive staining. Conclusion: Shock wave can promote the proliferation and differentiation of bone marrow mesenchymal stem cells.
