目的:建立实验动物牙周炎模型,评价含四环素的新型可注射牙槽骨修复材料用于引导牙周组织再生的实际效果。方法:选用15只雄性、健康的杂交犬,在双侧上颌尖牙的近中根面放置不锈钢网6周,建立实验动物牙周炎模型。在上颌尖牙近中牙槽骨处制作3壁骨内袋缺损,分别随机将含有1%、5%、10%四环素(TTC)的不饱和聚磷酸酯/β-磷酸三钙复合物(UPPE/β-TCP)直接注射入一侧的牙周骨缺损区(实验组),另一侧不植入任何材料(空白对照组)。术后16周处死动物,常规制备组织病理学样本,对实验组和空白对照组牙周组织的再生情况进行组织学和形态学评价,并对实验结果进行单因素方差分析和组间多重比较。结果:植入UPPE/β-TCP/TTC复合物的牙周骨缺损区内均可见新骨和新牙骨质形成等牙周组织再生现象,无明显的炎症反应,部分UPPE/β-TCP/TTC复合材料被新生骨组织所替代,且新生骨组织、新生牙骨质和新生牙周结缔组织附着宽度与空白对照组间的差异均有统计学意义(P<0.05)。结论:含TTC的新型可注射牙槽骨修复材料UPPE/β-TCP复合物可为牙周骨缺损区的愈合提供稳定的空间,并促进牙周组织的愈合,有望用于牙周组织再生治疗。
OBJECTIVE: To establish a model of periodontitis in experimental animals and evaluate the practical effect of a new type of injectable alveolar bone repair material containing tetracycline for guiding periodontal tissue regeneration. Methods: Fifteen male and healthy crossbred dogs were used to establish a model of periodontitis in experimental animals by placing a stainless steel mesh on the mesial root of bilateral maxillary canines. Three kinds of intraosseous defect were made in the maxillary canine near alveolar bone. The polyphosphate / β-tricalcium phosphate complex (UPPE) containing 1%, 5% and 10% tetracycline (TTC) / β-TCP) was injected directly into the periodontal bone defect (experimental group) on one side without implanting any material on the other side (blank control group). The animals were sacrificed 16 weeks after operation. Histopathology specimens were routinely prepared. The histopathological and morphological changes of the periodontal tissues in the experimental group and the blank control group were evaluated. One-way analysis of variance (ANOVA) and multiple comparisons among groups were performed. RESULTS: Periodontal tissue regeneration such as new bone formation and new cementum formation were observed in the periodontal defect with UPPE / β-TCP / TTC composite implantation. There was no obvious inflammatory reaction. Some UPPE / β-TCP / TTC composite material was replaced by new bone tissue, and there was significant difference (P <0.05) between the attachment width of fresh bone tissue, newborn cementum and newborn periodontal connective tissue and blank control group. CONCLUSION: The new injectable alveolar bone repair material UPTC / β-TCP containing TTC can provide a stable space for the healing of periodontal bone defects and promote the healing of periodontal tissues and is expected to be used in periodontal tissue regeneration therapy .
