目的:建立一种大鼠脂肪细胞抵抗模型原代培养方法,为科研实验提供原代胰岛素抵抗大鼠脂肪细胞。方法:取4周龄大鼠附睾的脂肪组织,原代培养前脂肪细胞经分化后,采用地塞米松造模方法,建立胰岛素抵抗的原代大鼠脂肪细胞模型。结果:原代前脂肪细胞培养后,经分化成为成熟的脂肪细胞,脂肪细胞内脂滴聚集,用油红O染色后,确定分化成为脂肪细胞。用地塞米松造模,用葡萄糖氧化酶法测定葡萄糖含量,计算各组细胞的葡萄糖消耗量,确定胰岛素抵抗脂肪细胞模型。结论:用4周龄大鼠附睾组织能培养出大鼠脂肪细胞,并用1 μmol/L浓度的地塞米松培养48 h造模法建立了胰岛素抵抗的脂肪细胞模型。
OBJECTIVE: To establish a primary culture method of rat adipocyte resistance model and provide primary insulin resistant rat adipocytes for scientific experiments. Methods: Adipose tissue of epididymis of 4-week-old rats was used to differentiate the primary cultured preadipocytes. Dexamethasone was used to establish the primary rat adipocyte model of insulin resistance. RESULTS: Primary cultured preadipocytes were differentiated into mature adipocytes. Lipid droplets were accumulated in adipocytes, stained with Oil Red O, and differentiated into adipocytes. Dexamethasone was used to make model. Glucose oxidase method was used to determine the glucose content. The glucose consumption of each group was calculated to determine the insulin resistance adipocyte model. CONCLUSION: Rat adipocytes can be cultured in epididymal tissue of 4-week-old rats and adipocyte models of insulin resistance have been established by 48 h culture with 1 μmol / L dexamethasone.
