目的研究红参提取物(HSI)对人脐静脉内皮细胞分泌一氧化氮(NO)、内皮素-1(ET-1)与6-酮前列腺素F_(1α)(6-K-PGF_(1α))的影响。方法体外培养人脐静脉内皮细胞株EA.hy926,随机分为5组:空白组、对照组、3个剂量(40,20,10 mL·L-1)实验组,将细胞传代培养后,分别用格里斯(Griess)试剂盒检测细胞内外的NO含量;用酶联免疫吸附测定法检测细胞上清液中的ET-1、内皮型一氧化氮合酶(eNOS)、环氧化酶-1(COX-1)以及6-酮-前列腺素F_(1α)(6-K-PGF_(1α))的含量。结果小中2个剂量HSI均可显著增加细胞内外的NO含量(P<0.05),3个剂量均可提高eNOS表达水平(P<0.05);大中2个剂量组可以减少细胞上清液中ET-1含量(P<0.05),而对COX-1以及6-K-PGF_(1α)的含量无明显影响(P>0.05)。结论 HSI可通过促进NO合成与释放,降低ET-1含量,发挥舒张血管作用。
Objective To study the effects of red ginseng extract (HSI) on the secretion of nitric oxide (NO), endothelin-1 (ET-1) and 6-ketoprostaglandin F 1alpha (6-K-PGF 1α ))Impact. Methods Human umbilical vein endothelial cell line EA.hy926 was cultured in vitro and randomly divided into 5 groups: blank group, control group and 3 doses (40, 20 and 10 mL·L-1) experimental groups. After subculture, The levels of NO in cells were detected by Griess kit. The levels of ET-1, eNOS and COX-1 in supernatants were detected by enzyme-linked immunosorbent assay (ELISA) (COX-1) and 6-keto-prostaglandin F (1α) (6-K-PGF_ (1α)). Results Both small and medium doses of HSI significantly increased intracellular and extracellular NO levels (P <0.05), and all three doses increased eNOS expression (P <0.05) ET-1 content (P <0.05), but had no significant effect on the content of COX-1 and 6-K-PGF1α (P> 0.05). Conclusion HSI can promote the synthesis and release of NO, reduce ET-1 content and exert vasodilatation.
