目的:研究消瘕方对人乳腺癌MDA-MB-435细胞株裸小鼠移植瘤中血管内皮生长因子(VEGF)表达的影响。方法:以体外培养的人乳腺癌MDA-MB-435细胞接种于裸小鼠右腋部位制造裸鼠皮下移植瘤模型,随机分为生理盐水组、5-氟尿嘧啶组及消瘕方小,中,大剂量组,每组小鼠予以相应药物治疗6周后脱颈处死,称瘤重,计算肿瘤生长抑制率,并采用RT-PCR法、免疫组化法检测肿瘤组织中VEGF mRNA及蛋白的表达。结果:消瘕方可抑制乳腺移植瘤生长,降低瘤体内VEGF mRNA表达(P<0.05),下调相应VEGF蛋白表达(P<0.05),其中以中剂量组效果更明显。结论:消瘕方的抑瘤作用与其下调瘤体内VEGF表达密切相关。
Objective: To study the effect of Xiaotan Recipe on the expression of vascular endothelial growth factor (VEGF) in nude mice transplanted with human breast cancer MDA-MB-435 cell line. METHODS: Human breast cancer MDA-MB-435 cells in vitro were inoculated into the right temporal region of nude mice to make a subcutaneous xenograft model of nude mice. The tumors were randomly divided into physiological saline group, 5-fluorouracil group, Xiao Xiao Fang, In the high-dose group, each group of mice was sacrificed after 6 weeks of drug treatment, and the tumor was weighed to calculate the tumor growth inhibition rate. RT-PCR and immunohistochemistry were used to detect the expression of VEGF mRNA and protein in the tumor tissue. . Results: Xiaotanfang can inhibit the growth of breast xenografts, reduce the expression of VEGF mRNA in tumors (P<0.05), and down-regulate the expression of VEGF protein (P<0.05). Among them, the middle dose group has more obvious effect. Conclusion: The anti-tumor effect of Xiaoyu Decoction is closely related to the down-regulation of VEGF expression in tumors.
