目的 :研究重组牛碱性成纤维细胞生长因子 (rbFGF)在视神经损伤修复中的作用。方法 :视神经部分损伤后 ,球后分别注射生理盐水、维生素B12 、bFGF ,伤后 4周测量闪光视神经诱发电位 (F VEP) ,进行轴突定量、视网膜神经节细胞 (RGCs)定量以及RGCs凋亡的检测 ,观察视神经损伤修复情况。结果 :伤后 4周时生理盐水和维生素B12 组对刺激无反应 ,bFGF组F VEP接近正常 ,80 0U、16 0 0U和 2 40 0UbFGF对RGCs挽救率分别为 2 4.5 %、2 7.3 %、2 8.5 % ,80 0U、16 0 0U和 2 40 0UbFGF组未发生溃变的轴突数分别是损伤未治疗组的 2 .0 3、2 .43、2 .31倍。流式细胞仪检测结果显示 ,bFGF治疗 7d后 ,RGCs凋亡率显著减少。结论 :bFGF可挽救视网膜神经节细胞的存活 ,减少轴突溃变 ,有抗凋亡作用 ,对视神经损伤有显著地促功能修复作用
Objective: To study the role of recombinant bovine basic fibroblast growth factor (rbFGF) in the repair of optic nerve injury. Methods: After partial optic nerve injury, normal saline and vitamin B12 and bFGF were injected into the posterior hemisphere respectively. The optic nerve evoked potential (F VEP) was measured 4 weeks after the injury and axon quantification, retinal ganglion cells (RGCs) quantification and RGCs apoptosis The detection of optic nerve injury repair situation. Results: At 4 weeks after injury, the saline and vitamin B12 groups did not respond to stimulation, and the FVEP in bFGF group was close to normal. The rescue rates of RGCs at 80 0U, 160 0U and 240 UUFGF were 24.5%, 2 7.3%, 2 The numbers of axons that failed to degenerate in 8.5%, 80%, 16% and 240% UbFGF groups were 2.03, 2.43 and 2.31 times, respectively, in the untreated group. Flow cytometry showed that after 7 days of bFGF treatment, the apoptotic rate of RGCs was significantly reduced. Conclusion: bFGF can save the survival of retinal ganglion cells, reduce axonal degeneration, have anti-apoptotic effects, and significantly promote the repair of optic nerve damage.
