目的:研究探讨复方藤梨根制剂抑制胃癌细胞转移的机制。方法:以SGC-7901胃癌细胞株建立胃癌裸鼠原位移植瘤模型,将裸鼠随机分为生理盐水组,复方藤梨根制剂低剂量、中剂量及高剂量组,接受连续21天口服灌注治疗。根据胃癌细胞转移的部位进行赋值评分,观察复方藤梨根制剂对裸鼠胃癌移植瘤转移的影响,并用荧光定量PCR仪(FQ-PCR)检测胃癌细胞乙酰肝素酶mRNA表达的影响。结果:根据胃癌细胞转移部位的赋值评分,生理盐水组与低剂量治疗组评分显著高于中、高剂量组;经FQ-PCR法检测,生理盐水组与低剂量治疗组的胃癌组织乙酰肝素酶mRNA表达显著高于中、高剂量治疗组。结论:在裸鼠动物模型,复方藤梨根制剂可能是通过下调乙酰肝素酶mRNA表达来抑制胃癌细胞的远处转移,发挥其抗肿瘤作用。
Objective: To investigate the mechanism of compound ramie root preparations inhibiting gastric cancer cell metastasis. Methods: Nude mice model of orthotopic transplanted gastric cancer was established by SGC-7901 gastric cancer cell line. The nude mice were randomly divided into normal saline group, low dose, middle dose and high dose group, treatment. According to the site of gastric cancer metastasis, we evaluated the effect of Fufang Tengligen on the metastasis of gastric cancer in nude mice. The expression of heparanase mRNA in gastric cancer cells was detected by FQ-PCR. Results: According to the assignment score of gastric cancer metastasis, the score of the saline group and the low-dose treatment group was significantly higher than that of the middle-dose and high-dose groups. The FQ-PCR assay showed that heparanase Enzyme mRNA expression was significantly higher than the medium and high dose treatment group. Conclusion: In the nude mice model, compound ramie root preparations may inhibit the distant metastasis of gastric cancer cells by down-regulating the expression of heparanase mRNA to exert its anti-tumor effect.
