最近研究发现,大部分淋巴瘤及白血病降钙素基因5'端可发生高度甲基化。异常的DNA片段经HPaⅡ消化后可以测到。笔者建立了PCR法可敏感地检测各种恶性血液病降钙素基因的高度甲基化,用以阐明PCR半定量甲基化的量与存在的白血病细胞数之间的关系。结果提示,该方法能有效地监测白血病细胞的变化,并可作为微小残留病检测的手段。
Recent studies have found that the majority of lymphomas and leukemic calcitonin gene 5’-end can be highly methylated. Abnormal DNA fragments were detected after digestion with HPaII. The authors established a PCR method that can sensitively detect the hypermethylation of calcitonin genes in various hematological malignancies to elucidate the relationship between the amount of semi-quantitative methylation of PCR and the number of leukemia cells present. The results suggest that this method can effectively monitor the changes of leukemia cells and can be used as a means of detection of minimal residual disease.
