目的通过短发夹RNA(shRNA)干扰沉默UbcH10基因的表达,并观察其对裸鼠移植结直肠癌的抑制作用。方法构建RNA干扰表达载体pGPU6/GFP/Neo/UbcH10-RNAi(pUbcH10-RNAi),建立UbcH10基因沉默细胞系HT-29/UbcH10-RNAi;采用蛋白印迹法(western blot)评价pUbcH10-RNAi对UbcH10基因表达的影响,观察移植结直肠癌裸鼠的肿瘤生长情况。结果 HT-29/UbcH10-RNAi细胞中UbcH10基因表达明显下调;与对照组比较HT-29/UbcH10-RNAi组肿瘤生长明显减慢,在接种肿瘤细胞12 d后隐约可见瘤体,36 d后断颈处死剥离瘤体重量仅是对照组肿瘤的40%左右。结论短发夹RNA(shRNA)沉默UbcH10基因对裸鼠移植性大肠癌有抑制作用,提示抑制UbcH10基因表达可能是大肠癌治疗的潜在方法。
Objective To silence the expression of UbcH10 gene by short hairpin RNA (shRNA) and observe its inhibitory effect on colorectal cancer in nude mice. RNA interference expression vector pGPU6 / GFP / Neo / UbcH10-RNAi (pUbcH10-RNAi) was constructed and the UbcH10 gene silencing cell line HT-29 / UbcH10-RNAi was established. Western blotting was used to evaluate the effect of pUbcH10- Expression of the impact of observation of tumor growth in transplanted colorectal cancer nude mice. Results The expression of UbcH10 gene was down-regulated in HT-29 / UbcH10-RNAi cells. Compared with the control group, the tumor growth of HT-29 / UbcH10-RNAi group was significantly slowed down. Cervical ablation of tumor weight was only about 40% of control tumors. Conclusion Short hairpin RNA (shRNA) silencing UbcH10 gene has inhibitory effect on transplanted colorectal cancer in nude mice, suggesting that inhibition of UbcH10 gene expression may be a potential therapeutic approach for colorectal cancer.
