地塞米松磷酸盐是一种肾上腺皮质激素类药物,在一定的温度,pH环境下,地塞米松磷酸钠会水解为难溶药物地塞米格,影响其稳定性。本论文确定了用HPLC法同时测定出地塞米松磷酸钠和其水解产物地塞米松的色谱条件;研究了药物载体羟丙基-β-环糊精(HP-β-CD)存在及不存在条件下地塞米松磷酸盐的水解行为.实验结果表明,HP-β-CD与水解产物地塞米松发生也接反应后能增加难溶药物地塞米松在水中的溶解度,同时破坏了水解反应的平衡,加速了地塞米松磷酸钠的水解作用。在反应温度60℃,pH为7.0时地塞米格磷酸钠的水解速率最小为0.007D-1.地塞米格磷酸钠水解产物的液和色谱图结果表明,其水解产物中除地塞米松产物外,还存在其它未知水解产物.
