目的观察消白合剂对人A375黑素瘤细胞生长、酪氨酸酶活性及黑素含量的影响。方法设消白合剂水提液体0.01、0.1、1、10g/L浓度实验组和空白对照组,体外观察对人A375黑素瘤细胞的增殖、酪氨酸酶的活性以及黑素含量的调节作用。四甲基偶氮唑蓝(MTT)比色法测定消白合剂对细胞增殖的影响,酶学方法测定对酪氨酸酶活性的影响,比色法(490 nm)测定对黑素含量的影响。结果消白合剂在0.01～0.1 g/L浓度范围内对细胞增殖、酪氨酸酶活性及黑素的合成有促进作用,而随着浓度的继续增高反而起抑制作用。结论消白合剂对人A375黑素瘤细胞株黑素的合成具有双向调节作用,临床运用时应该考虑到药物的量的优化。
Objective To observe the effect of Xiaobai Mixture on the growth, tyrosinase activity and melanin content of human A375 melanoma cells. Methods The concentrations of 0.01, 0.1, 1 and 10 g/L of Xiaobai Mixture were extracted and the control group was set up. The proliferation, tyrosinase activity and melanin content of human A375 melanoma cells were observed in vitro. . MTT colorimetric method was used to determine the effect of Xiaobai Mixture on cell proliferation, enzymatic method was used to determine the effect of tyrosinase activity, colorimetric method (490 nm) was used to determine melanin content. . Results Xiaobai Mixture promoted cell proliferation, tyrosinase activity, and synthesis of melanin in the concentration range of 0.01-0.1 g/L, but inhibited it with increasing concentration. [Conclusion] Xiaobai Mixture has two-way adjustment effect on melanin synthesis in human A375 melanoma cell line. The optimization of drug dosage should be considered in clinical application.
