目的:观察大鼠长期口服黄苓茎叶总黄酮(TF)产生的毒性反应及其严重程度,毒性反应的耙器官及其损害的可逆性,为确定人用安全剂量提供依据。方法:Wistar大鼠,分为0.5、1.0、2.0g·kg-1TF和CMC对照组,灌胃给药,连续90d,观察用TF90d和停药14d时对大鼠血液学、血液生化学、重要脏器系数及病理组织学的影响。结果:用药90d时2.0g·kg-1TF组大鼠RBC高于CMC对照组,停药14d恢复正常。其它组大鼠的各项指标均无异常改变,也未发现迟发性毒性反应。结论:本实验所用高剂量高达小鼠最大耐受量的1/5,除出现一过性胃肠道毒性反应及用药90d时RBC升高外,未见明显毒性反应,提示TF毒性较低
OBJECTIVE: To observe the toxicity and severity of long-term oral administration of total flavonoids (TFs) in Huangling and its reversibility in harming organs and toxicity, and provide the basis for the determination of safe dosage for human use. Methods: Wistar rats were divided into 0.5, 1.0, 2.0g · kg-1TF and CMC control groups. The rats were administered intragastrically for 90 days. The effects of TF90d and withdrawal for 14 days on the hematology, Blood biochemistry, vital organ coefficient and histopathology. Results: The RBC of 2.0g · kg-1 TF group was higher than that of CMC control group at 90 days after drug administration, and returned to normal after 14 days of drug withdrawal. Other groups of rats showed no abnormal changes in the indicators, nor delayed toxic reactions. Conclusion: In this experiment, the high dose up to 1/5 of the maximum tolerable dose of mice, in addition to a transient gastrointestinal toxicity and RBC increased 90 days after treatment, no significant toxicity, suggesting that TF toxicity is low
