采用 377DNA测序仪 (P .E .Corp .)用AFLP技术对 2 5种分别具有高产 /低产、抗白粉病 /感白粉病、抗寒 /不抗寒、死皮 /不死皮性状的橡胶树 (HeveabrasiliensisMull.Ary)无性系 (其中Wickham种质 15种 ,Amazon野生橡胶树种质 10种 )进行了指纹图谱分析。从 6 4对引物组合中选出 2对引物组合构建了所有被研究种质的指纹图谱。 2对引物共扩增出 5 18条谱带 ,多态性比率超过 98.6 %。遗传多样性分析表明 ,橡胶树种质间遗传距离为 0 .2 5~ 0 .81,RRIM6 0 0种质内遗传距离为 0 .0 7~ 0 .17。通过基因分型分析获得一条大小为 32 0bp的抗白粉病种质特有的片段。根据以上的AFLP数据进行了聚类分析 ,结果表明所有被研究种质几乎是依次聚成一个大类。
A total of 25 rubber tree species with high yield / low yield, powdery mildew / powdery mildew, cold / non-cold tolerant, dead skin / undeveloped skin were harvested by using 377 DNA sequencer (P.E.Corp.) .Ary) clones (including 15 Wickham germplasms and 10 species of Amazon wild rubber germplasm) were analyzed by fingerprinting. Two pairs of primer combinations were selected from 64 pairs of primers to construct a fingerprint of all the germplasm studied. A total of 5 18 bands were amplified with 2 pairs of primers, and the polymorphic ratio was over 98.6%. The analysis of genetic diversity showed that the genetic distance between rubber tree germplasm ranged from 0.52 to 0.81, and the RRIM60 genetic distance ranged from 0.07 to 0.17. A 3200 bp fragment unique to powdery mildew germplasm was obtained by genotyping analysis. The clustering analysis based on the above AFLP data showed that all the studied germplasms were clustered into almost one category in turn.
