目的探讨Wistar大鼠移植性CBRH-7919肝癌模型的制作方法。材料与方法先用CBRH-7919大鼠肝癌细胞瘤株接种于裸鼠肩胛部皮下,再将生成的肿瘤组织移植接种于42只成年Wistar大鼠肝脏内,术前1周、术后连续2周内给每只大鼠肌注地塞米松2.5mg/天。手术4周后用彩超和磁共振检查肿瘤生长情况,筛选建模成功的大鼠,并从中随机抽取5只大鼠,处死后取出肝脏内肿瘤进行病理学检查。结果 42只大鼠中有2只死亡。剩余的40只大鼠中有35只肝脏内共长出41个结节或肿块,成功率为87.5%。肿瘤直径约(7±3)mm,有1只于腹壁手术切口处同时长出一肿块,直径约6.5mm。病理类型均为肝细胞性肝癌。结论直视下向免疫抑制大鼠肝脏内接种制作大鼠肝细胞癌模型,是一种操作方便、简单、成功率高的方法。能很好满足于影像学诊断、介入治疗等实验方面的研究。
Objective To investigate the method of making a transplanted CBRH-7919 hepatocellular carcinoma model in Wistar rats. MATERIALS AND METHODS CBRH-7919 rat hepatoma cell line was inoculated subcutaneously in the scapular of nude mice. The resulting tumor tissues were transplanted in the liver of 42 adult Wistar rats. One week before surgery and two weeks after surgery Each rat was intramuscularly given dexamethasone 2.5 mg / day. After 4 weeks of surgery, the tumor growth was examined by color Doppler ultrasound and magnetic resonance imaging. The rats were screened successfully. Five rats were randomly selected from the tumors and the tumors in the liver were removed for pathological examination. Results Two out of 42 rats died. A total of 41 nodules or lumps were found in 35 of the remaining 40 rats with a success rate of 87.5%. Tumor diameter of about (7 ± 3) mm, there are 1 abdominal incision at the same time grow a mass, a diameter of about 6.5mm. Pathological types are hepatocellular carcinoma. Conclusions Under the direct vision, immunization of the liver of immunosuppressed rat inoculation production of rat hepatocellular carcinoma model is a convenient, simple, high success rate. Can be well satisfied with the imaging diagnosis, interventional treatment experiments.
