目的:建立晚期肺癌的动物模型,研究其对化疗药物及益气养阴清热方的治疗反应状况,寻找适合评价以控制症状、改善生活质量为目的疗法的实验模型。方法:通过胸腔移植的方法建立晚期肺癌的动物模型,研究该模型的生存期及对氟尿嘧啶(20mg/kg)、顺铂(3mg/kg)、益气养阴清热方浓缩中药溶液(28.6mg/kg)的治疗反应状况,用免疫组化的方法检测了肿瘤组织内表皮生长因子(EGF)和表皮生长因子受体(EGFR)等蛋白表达(400×),分析了该模型较皮下移植模型的优势。结果:经过治疗后,胸腔组内的中药组、顺铂组体重下降程度较对照组和氟尿嘧啶组体重下降程度低(P<0.05)。皮下组内各小组体重变化无明显差异。胸腔组较相应皮下组饲料消耗量少。胸腔组内的对照组和氟尿嘧啶组饲料消耗量较其他组明显减少(P<0.05),益气养阴清热方组、顺铂组间无明显差异。皮下组内各小组饲料消耗量无明显差异。对照组和氟尿嘧啶组中EGF、EGFR阳性细胞数较其他组明显增多。结论:胸腔移植模型可作为晚期肺癌动物模型,胸腔移植模型较皮下移植模型能更好地反映药物的作用,化疗药物、益气养阴清热方对EGF、EGFR等的表达有不同程度的抑制作用,益气养阴清热方对抑制肿瘤发展优势不明显,但对改善模型生存时间、生活状态有较好的作用。
OBJECTIVE: To establish an animal model of advanced lung cancer and study its response to chemotherapeutic drugs and Yiqi Yangyin Qingre prescription, and to find an experimental model suitable for the evaluation of therapies to control the symptoms and improve the quality of life. Methods: The animal model of advanced lung cancer was established by the method of thoracic transplantation. The survival of the model and the effects of fluorouracil (20mg / kg), cisplatin (3mg / kg) kg). The protein expression of epidermal growth factor (EGF) and epidermal growth factor receptor (EGFR) in the tumor tissue was detected by immunohistochemistry (400 ×), and the expression of EGFR, Advantage. Results: After treatment, the body weight loss in the thoracic cavity group was lower than that in the control group and fluorouracil group (P <0.05). There was no significant difference in body weight among subcutaneous groups. Chest group than the corresponding subcutaneous feed consumption less. There was no significant difference between the control group and the fluorouracil group in the Yiqi Yangyin Qingre Decoction group and the cisplatin group. Subcutaneous group within each group of feed consumption no significant difference. The number of EGF and EGFR positive cells in control group and fluorouracil group was significantly higher than that in other groups. Conclusion: The model of thoracic cavity transplantation can be used as an animal model of advanced lung cancer. The model of thoracic cavity transplantation can better reflect the effect of drugs than the model of subcutaneous transplantation. Chemotherapy drugs and Yiqi Yangyin Qingre prescription can inhibit the expression of EGF and EGFR to some extent , Yiqi Yangyin Qingrefang has no obvious advantage in inhibiting tumor development, but it has a good effect on improving model survival time and living condition.
