目的探讨Ln5γ2对胃癌SGC-7901细胞迁移、侵袭的影响及可能机制。方法实验分为对照组、Ln5γ2趋化组、上皮生长因子受体(epithelial growth factor receptor,EGFR)干扰组、空载体组。以Ln5γ2诱导趋化SGC-7901,观察穿过铺和未铺Matrigl胶的Transwell小室滤膜的细胞数目。采用ELISA和免疫荧光方法检测细胞缓冲液中MMP-2和MMP-9浓度以及细胞骨架结构改变。结果 Ln5γ2诱导趋化SGC-7901,穿过铺和未铺Matrigl胶的Transwell小室滤膜的细胞数目明显高于对照组(P<0.01),EGFR干扰后数目与对照组无显著差异(P>0.05)。EGFR干扰组细胞MMP-2和MMP-9浓度下降,细胞骨架结构模糊,伪足形成减少或消失。结论 Ln5γ2可以促进胃癌SGC-7901侵袭迁移,可能机制为通过EGFR改变细胞骨架结构,影响MMPs的分泌。
Objective To investigate the effect of Ln5γ2 on the migration and invasion of gastric cancer SGC-7901 cells and its possible mechanism. Methods The experiment was divided into control group, Ln5γ2 chemotactic group, epithelial growth factor receptor (EGFR) interference group and empty vector group. The chemotaxis of SGC-7901 was induced by Ln5γ2 and the number of cells passing through the Transwell chamber membrane coated with Matrigl glue was observed. The concentration of MMP-2 and MMP-9 in cell buffer and the change of cytoskeleton structure were detected by ELISA and immunofluorescence. Results The number of cells transfected with SGC-7901 and transfected with Transwell chamber membrane coated with Matrigl glue by Ln5γ2 was significantly higher than that of the control group (P <0.01). There was no significant difference between EGFR group and control group (P> 0.05 ). EGFR interference group decreased the concentration of MMP-2 and MMP-9, cytoskeleton structure fuzzy, pseudopodia decreased or disappeared. Conclusion Ln5γ2 can promote the invasion and migration of gastric cancer cell line SGC-7901. The possible mechanism is that EGFR can change the cytoskeleton structure and affect the secretion of MMPs.
