目的 :探讨原癌基因WNT1、β-连环蛋白(β-catenin)、T细胞因子4(TCF4)在基底细胞癌(BCC)的表达模式及意义。方法:采用免疫组化方法、免疫印迹及实时定量PCR对62例BCC和56例脂溢性角化病(SK)的皮损组织标本进行WNT1、β-catenin、TCF4基因m RNA及蛋白的表达分析。结果 :WNT1、β-catenin、TCF4在BCC中的阳性率分别为87.10%(54/62)、83.87%(52/62)、93.55%(58/62)高于SK中的的25.00%(14/56)、19.64%(11/56)、10.71%(6/56),差异具有统计学意义(P值分别为9.35×10~(-12),2.86×10~(-12),1.91×10~(-12));WNT1、β-catenin、TCF4基因m RNA及蛋白在BCC中表达升高,差异具有统计学意义(P值分别为0.018,0.027,0.038)。结论:WNT1、β-catenin、TCF4异常表达模式可能是促进BCC发生的重要因素,与BCC的发生密切相关。
OBJECTIVE: To investigate the expression patterns and significance of the oncogenes WNT1, β-catenin and TCF4 in basal cell carcinoma (BCC). Methods: The expression of m RNA and protein of WNT1, β-catenin and TCF4 genes in 62 cases of BCC and 56 cases of seborrheic keratosis (SK) were detected by immunohistochemistry, immunoblot and real-time quantitative PCR analysis. Results: The positive rates of WNT1, β-catenin and TCF4 in BCC were 87.10% (54/62), 83.87% (52/62), 93.55% (58/62) /56)(19.64%(11/56),10.71%(6/56)), with statistical significance (P values were 9.35 × 10 ~ (-12), 2.86 × 10 ~ (-12), 1.91 × 10 ~ (-12)). The expression of m RNA and protein of WNT1, β-catenin and TCF4 increased in BCC, the difference was statistically significant (P = 0.018,0.027,0.038). Conclusion: The abnormal expression pattern of WNT1, β-catenin and TCF4 may play an important role in promoting the occurrence of BCC, which is closely related to the occurrence of BCC.
