黄芪对HSC-T6细胞Col-I、TIMP1mRNA影响表达的研究

观察黄芪对HSC—T6细胞I型胶原 (Col-I)、基质金属蛋白酶组织抑制因子 1(TIMP1)mRNA表达的影响。用黄芪 0 .5mg/ml、1.0mg/ml浓度作用于HSC—T6细胞 4 8h ,以逆转录定量PCR方法测定其对HSC—T6细胞Col -I、TIMP1mRNA表达的影响。提示黄芪 0 .5mg/ml、1.0mg/ml浓度HSC—T6细胞Col-ImRNA表达水平为 2 .5± 0 .71、1.5±0 .0 1,与空白对照组比较 ,具有显著性差异 ;TIMP1mRNA表达水平依次为 4 .0 0± 0 .0 1、4 .0 0± 1.4 1,与空白对照组比较 ,无显著性差异。指出黄芪可明显降低HSC—T6细胞Col-ImRNA表达水平 ,而对TIMP1mRNA表达水平无明显影响。 The effects of astragalus on the expression of type I collagen (Col-I) and matrix metalloproteinase inhibitor 1 (TIMP1) mRNA in HSC-T6 cells were observed. HSC-T6 cells were treated with astragalus at the concentration of 0.5 mg/ml and 1.0 mg/ml for 48 hours. The effect of HSC-T6 cells on expression of Col-I and TIMP1 mRNA was determined by reverse transcription quantitative PCR. It was suggested that the expression of Col-I mRNA in HSC-T6 cells at the concentration of 0.5 mg/ml and 1.0 mg/ml was 2. 5± 0 .71 and 1.5±0. 0 1, respectively. Compared with the blank control group, there was a significant difference; TIMP 1 mRNA The expression levels were 4.00±0.10,4.00±1.41 in turn, compared with the blank control group, there was no significant difference. It was pointed out that Astragalus membranaceus could significantly reduce the expression of Col-I mRNA in HSC-T6 cells, but had no effect on the expression of TIMP1 mRNA.
