TB-Chen病毒是一株分离自中国胃肠炎患儿的A组轮状病毒。研究结果表明,TB-Chen病毒的NSP5和NSP6由第10基因组节段编码,该节段全长816bp。NSP5含200个氨基酸,由第22~624位核苷酸(nt)编码,预测分子量为21.9 kD,等电点为7.86。NSP6含92个氨基酸,由第80~355位nt编码,预测分子量为11 kD,等电点为9.65。对编码NSP5和NSP6的ORF核苷酸序列的分子进化分析结果表明,A组RV的NSP5至少可分为7个不同的基因型(H1~H7型),TB-Chen株NSP5属于H2型;NSP6至少可分为8个基因型(h1~h8型),TB-Chen株NSP6属于h2型。这是首个对A组人RV的NSP6基因分型的研究结果的报道,并且我们建议NSP6的基因型以英文字母“h”代表,如TB-Chen株的NSP5/NSP6基因型为H2h2,69M株的NSP5/NSP6基因型为H7h7,Wa株与KU株的NSP5/NSP6基因型为H1h8。
The TB-Chen virus is a group A rotavirus isolated from children with gastroenteritis in China. The results showed that the TB-Chen virus NSP5 and NSP6 encoded by the tenth genome segment, the full length of 816bp. NSP5 contains 200 amino acids and encodes nucleotides 22 to 624 (nt), with a predicted molecular mass of 21.9 kD and an isoelectric point of 7.86. NSP6 contains 92 amino acids and encodes nt 80-355, with a predicted molecular weight of 11 kD and an isoelectric point of 9.65. The results of molecular phylogenetic analysis of ORF nucleotide sequences encoding NSP5 and NSP6 showed that NSP5 of group A RV can be divided into at least 7 different genotypes (H1 to H7), NSP5 of TB-Chen strain belongs to H2 type, NSP6 It can be divided into at least eight genotypes (h1 ~ h8), and TB-Chen strain NSP6 belongs to h2 type. This is the first report of a study of NSP6 genotyping of human RV in group A, and we suggest that the genotype of NSP6 is represented by the English letter “h ” such as the NSP5 / NSP6 genotype of TB-Chen strain is H2h2 , The NSP5 / NSP6 genotype of 69M strain is H7h7, and the NSP5 / NSP6 genotype of Wa strain and KU strain is H1h8.
