目的 探讨长期苯那普利 ( benazepril)治疗逆转高血压左心室肥厚的作用 ,其量效关系及与局部肾素 -血管紧张素系统 ( RAS)的关系。方法 10周龄的雄性自发性高血压大鼠 ( SHR)给予三个不同剂量的苯那普利 ( 3、 10、30 mg.kg- 1· d- 1 )口服 16周 ,性别、年龄配对的 SHR和 Wistar- Kyoto( WKY)大鼠作为对照。测量血压、体重、左心室肥厚的指标 ,采用放射免疫方法测定左心室组织的血管紧张素 ( Ang )浓度。结果 苯那普利治疗组 SHR的SBP和 L VM/ BW比显著低于对照组 SHR,具有量效关系 ;治疗组心肌组织中的 Ang 浓度显著低于未治疗组。结论 苯那普利长期治疗可减轻或逆转 SHR大鼠左心室肥厚的发展 ,具有量效关系 ,并与局部 RAS相关
Objective To investigate the effect of long-term benazepril treatment on reversing left ventricular hypertrophy in patients with hypertension and its relationship with the dose-response relationship and local renin-angiotensin system (RAS). Methods Ten-week-old male spontaneously hypertensive rats (SHR) were given benazepril (3, 10, 30 mg.kg-1 · d-1) orally at 16 different doses for three different doses SHR and Wistar-Kyoto (WKY) rats served as controls. The indexes of blood pressure, body weight and left ventricular hypertrophy were measured. The concentration of angiotensin (Ang) in left ventricular tissue was measured by radioimmunoassay. Results The SBP and L VM / BW ratios of SHR in benazepril treatment group were significantly lower than those in control group, with a dose-response relationship. The concentration of Ang-2 in myocardial tissue of treatment group was significantly lower than that of untreated group. Conclusion Long-term benazepril treatment can reduce or reverse the development of left ventricular hypertrophy in SHR rats, with dose-response relationship and local RAS
