为了解近几年河南地区猪圆环病毒2型(PCV2)的遗传进化情况,本研究对采自2011年~2015年河南省13个地区的28份疑似患断奶仔猪多系统衰竭综合征(PMWS)猪的病料样品进行PCR检测,并将检出的阳性病料样品接种无PCV的PK-15细胞,分离PCV2,进而用IPMA进行鉴定。最后对所获分离株进行全基因组克隆和序列分析。结果显示,28份样品中阳性率为75%(21/28),并成功分离出21株PCV2,其全基因序列同源性为95.4%~99.9%;在遗传演化关系上,21株PCV2分离株分布于两个基因型(PCV2b和PCV2d),16株属于PCV2d,而其余5株属于PCV2b基因型,这也说明PCV2d逐渐成为河南地区的优势流行株。此外,通过对其Cap蛋白氨基酸序列的预测与分析,发现其主要抗原区存在12处位点明显变异,这可能影响其抗原性及免疫原性。本研究为河南地区PCV2的防控提供了依据。
In order to understand the genetic evolution of porcine circovirus type 2 (PCV2) in Henan Province in recent years, this study collected 28 suspected swine SSR (PMWS) from 13 areas in Henan Province from 2011 to 2015, ) Swine samples were tested by PCR, and the positive samples detected were inoculated with PCV-free PK-15 cells, PCV2 was isolated and further identified by IPMA. Finally, the isolates obtained by genome-wide cloning and sequence analysis. The results showed that the positive rate was 75% (21/28) in 28 samples and 21 strains of PCV2 were successfully isolated. The full-length sequence of PCV2 was 95.4% -99.9%. In the relationship between genetic evolution, 21 PCV2 isolates Strain distribution in two genotypes (PCV2b and PCV2d), 16 strains belong to PCV2d, and the remaining 5 strains belong to PCV2b genotypes, which also shows that PCV2d has gradually become the dominant epidemic strain in Henan. In addition, by predicting and analyzing the amino acid sequence of its Cap protein, it was found that there were 12 significant variations in its major antigenic region, which may affect its antigenicity and immunogenicity. This study provides a basis for the prevention and control of PCV2 in Henan Province.
