目的研究医用胶原蛋白调节TNF-α治疗变应性接触性皮炎的作用及机制。方法制造二硝基氟苯致小鼠变应性接触性皮炎模型,医用胶原蛋白连续14 d涂抹动物皮肤,对耳部组织进行病理学检查,应用ELISA方法测试TNF-α、IL-2等炎症介质。结果与攻击前比较,各组小鼠在攻击后12、24 h均出现十分明显的耳变态反应(P<0.01);与空白对照组比较,医用胶原蛋白低剂量组小鼠在攻击后12 h时耳变态反应评分升高(P<0.05),在攻击后24 h时模型对照组耳变态反应评分明显升高(P<0.05);与模型对照组比较,在攻击后24 h时医用胶原蛋白低、中和高剂量组小鼠的耳变态反应评分均降低(P<0.05)。与空白对照组比较,模型对照组、地塞米松对照组和医用胶原蛋白低、中、高剂量组小鼠右耳片的炎症细胞数明显地增加(P<0.01);与模型对照组比较,地塞米松对照组和医用胶原蛋白低、高剂量组小鼠右耳片的炎症细胞数减少(P<0.05或P<0.01)。与空白对照组比较,模型对照组、地塞米松对照组和医用胶原蛋白中、高剂量组小鼠左右耳片厚度差明显地变大(P<0.01)。与模型对照组比较,医用胶原蛋白低剂量组小鼠左右耳片厚度差变小(P<0.05);与空白对照组比较,模型对照组和医用胶原蛋白低剂量组小鼠血清中炎症因子TNF-α含量明显地升高(P<0.01);与模型对照组比较,地塞米松对照组小鼠血清中炎症因子TNF-α含量明显地降低(P<0.01),医用胶原蛋白高剂量组小鼠血清中炎症因子TNF-α含量明显降低(P<0.05);对动物体重没有明显影响。结论医用胶原蛋白对变应性接触性皮炎有明显的治疗作用,可能通过调节血清中TNF-α进而减轻耳部炎性反应有关。
Objective To study the effect and mechanism of medical collagen in regulating the treatment of allergic contact dermatitis with TNF-α. Methods The mouse model of allergic contact dermatitis induced by dinitrofluorobenzene was established. Compared with the model control group, the difference of the thickness of left and right ears of the mice in the low-dose medical collagen group was smaller (P <0.05). Compared with the blank control group, the serum levels of the inflammatory cytokines TNF (P <0.01). Compared with the model control group, the level of TNF-α in the serum of mice in the dexamethasone control group was significantly decreased (P <0.01), the high dose of the medical collagen group was smaller The content of TNF-αin the serum of the rats decreased obviously (P <0.05), and had no significant effect on the weight of the animals. Conclusion Medical collagen has obvious therapeutic effect on allergic contact dermatitis, which may be related to the reduction of inflammatory reaction in the ear by regulating TNF-α in serum.
