目的:观察大鼠同种异体穿透性角膜移植后无干预下角膜新生血管的生长演变。方法:SPF级雌性SD大鼠34眼行同种异体穿透性角膜移植,术后4,7,15和30d在显微镜下观察角膜新生血管的生长情况,通过公式C/12×3.14×[r2-(r-I)2]计算角膜新生血管的面积。结果:在移植的34眼中出现新生血管的29眼(85%)。新生血管最初环角膜缘呈毛刷状生长,后向角膜中央蔓延,为扭曲粗大的血管,末端呈树枝状分支。高峰时期交叉密布成网状,以后逐渐萎缩。角膜移植后4,7,15和30d角膜新生血管平均面积分别为11.8±3.5mm2,18.5±4.0mm2,14.4±4.3mm2和6.0±1.8mm2,总体平均面积为12.7±1.9mm2;新生血管面积占角膜面积百分比值分别为30.8%±8.7%,65.3%±12.8%, 59.4%±14.5%和36.2%±10.9%,总体为48.7%±6.4%。结论:大鼠同种异体穿透性角膜移植后角膜新生血管在第4d时出现,7d时面积达最大值;以后逐渐下降,30d时约降至7d时面积的1/2。
Objective: To observe the growth and development of corneal neovascularization after allogeneic penetrating keratoplasty in rats. Methods: 34 eyes of SPF female Sprague-Dawley rats received allogenic penetrating keratoplasty. The growth of corneal neovascularization was observed under microscope 4, 7, 15 and 30 days after operation. The formula of C / 12 × 3.14 × [r2 - (rI) 2] Calculate the area of corneal neovascularization. RESULTS: Thirty-nine eyes (85%) of neovascularized vessels were found in the transplanted 34 eyes. The neovascularization initially showed a brush-like growth of the corneal limbus, spreading toward the central cornea, distorting the thick blood vessels and ending with a dendritic branch. The peak period of cross-shaped into a mesh, then gradually shrinking. The average area of corneal neovascularization at 4, 7, 15 and 30 days after corneal transplantation were 11.8 ± 3.5mm2, 18.5 ± 4.0mm2, 14.4 ± 4.3mm2 and 6.0 ± 1.8mm2, respectively. The average area of corneal neovascularization was 12.7 ± 1.9mm2. Corneal area percentages were 30.8% ± 8.7%, 65.3% ± 12.8%, 59.4% ± 14.5% and 36.2% ± 10.9%, respectively, with an overall 48.7% ± 6.4%. CONCLUSION: Corneal neovascularization occurs on day 4 after allogeneic penetrating keratoplasty in rats. The area reached the maximum on the 7th day and gradually decreased after that, and dropped to about 1/2 of that on the 7th day after 30 days.
