目的:研究西黄丸乙醇提取物对荷瘤大鼠的抗肿瘤作用及其免疫学机制。方法:采用w256乳腺癌细胞建立大鼠荷瘤模型,随机分为7组:正常组,模型对照组,香菇多糖组,西黄丸高剂量组,西黄丸乙醇提取物低、中、高剂量组。治疗14?d,取瘤,计算抑瘤率;采用流式细胞技术检测外周血CD4+、CD8+和B7-1的含量;采用ELISA技术检测外周血中IL-6、IL-10、TGF-β的表达水平,结果与模型对照组和香菇多糖组比较。结果:西黄丸乙醇提取物高剂量对荷瘤大鼠的肿瘤抑制率为36.8%,与模型组对比西黄丸乙醇提取物可明显提高CD4+、CD8+细胞的比例,降低IL-6、IL-10、TGF-β的表达水平(P<0.05);相当原药剂量的乙醇提取物优于西黄丸原制剂。结论:西黄丸乙醇提取物提高了原制剂的生物利用度,具有明显抑制肿瘤的生长的作用,改善淋巴细胞亚群比例,调节肿瘤相关炎性因子表达。
Objective: To study the anti-tumor effect of Xihuangwan ethanol extract on tumor-bearing rats and its immunological mechanism. Methods: W256 breast cancer cells were used to establish tumor-bearing model in rats and were randomly divided into 7 groups: normal group, model control group, lentinan, high-dose Xixi Wan group, low, medium and high dose of ethanol extract group. The tumors were treated for 14 days and the tumor inhibition rate was calculated. The levels of CD4 +, CD8 + and B7-1 in peripheral blood were detected by flow cytometry. The levels of IL-6, IL-10 and TGF-β The expression level was compared with the model control group and lentinan group. Results: Compared with model group, Xihuang Wan ethanol extract could significantly increase the proportion of CD4 +, CD8 + cells and decrease the levels of IL-6, IL-6 and IL-6 in tumor-bearing rats. 10, TGF-β expression level (P <0.05); the equivalent of the original dose of ethanol extract is better than Xihuang Wan original preparation. Conclusion: XWG ethanol extract can improve the bioavailability of the original preparation, inhibit the growth of the tumor, improve the proportion of lymphocyte subsets and regulate the expression of tumor-associated inflammatory factors.
