目的:探讨改构苹果多糖(modified apple polysaccharides,MAP)对人结肠癌HT29细胞凋亡的影响及其机制。方法:采用MTT法观察不同浓度MAP对HT29细胞增殖的影响;TUNEL法和流式细胞术研究MAP对HT29细胞凋亡的作用;Western Blot法检测给予不同浓度MAP后,HT29细胞中凋亡相关蛋白caspase3,8,9、Bax和Bcl-2表达的变化。结果:与对照组相比,MAP对HT29细胞增殖有明显的抑制作用且能诱导HT29细胞的凋亡;MAP可促进HT29细胞caspase3,8,9及Bax的表达,降低Bcl-2的表达,且均呈剂量依赖性。结论:MAP可以通过内部和外部途径诱导HT29细胞凋亡。
Objective: To investigate the effect of modified apple polysaccharides (MAP) on the apoptosis of human colon carcinoma HT29 cells and its mechanism. Methods: MTT assay was used to observe the effect of different concentrations of MAP on the proliferation of HT29 cells. TUNEL and flow cytometry were used to study the effect of MAP on apoptosis of HT29 cells. Western blot was used to detect the expression of apoptosis-related protein Caspase3, 8, 9, Bax and Bcl-2 expression. Results: Compared with the control group, MAP significantly inhibited the proliferation of HT29 cells and induced the apoptosis of HT29 cells. MAP promoted the expression of caspase3, 8, 9 and Bax and decreased the expression of Bcl-2 in HT29 cells All in a dose-dependent manner. Conclusion: MAP can induce HT29 cell apoptosis through both internal and external pathways.
