【目的】探索生物信息学方法在Reelin剪切蛋白查找中的应用,并筛选大脑皮质中可能对Reelin有剪切作用的蛋白。【方法】以ADAMTS-4蛋白为基准,利用生物信息学方法,以功能域查找相似功能蛋白,并结合同源性比对方法从中筛选出剪切Reelin的候选蛋白,并对候选蛋白的理化性质、亚细胞定位、功能域、二级结构和组织定位进行分析。【结果】ADAMTS-2、ADAMTS-3、ADAMTS-10、ADAMTS-16与ADAMTS-4有相似的理化性质,且均属于分泌蛋白,在大脑皮质中可能有分布。ADAMTS-2、ADAMTS-3、ADAMTS-10、ADAMTS-16包含ADAMTS-4的功能域,且功能域内的二级结构相似。【结论】筛选到了可能对Reelin有剪切作用的蛋白ADAMTS-2、ADAMTS-3、ADAMTS-10和ADAMTS-16。
【Objective】 To explore the application of bioinformatics in the search of Reelin cutin and to screen the proteins in the cerebral cortex that may have effects on Reelin. 【Method】 Based on the ADAMTS-4 protein, bioinformatics methods were used to search similar functional proteins by functional domains. The candidate proteins of cut Reelin were screened out by homology comparison method. The physical and chemical properties of the candidate proteins , Subcellular localization, functional domains, secondary structure and tissue localization. 【Results】 ADAMTS-2, ADAMTS-3, ADAMTS-10 and ADAMTS-16 shared similar physicochemical properties with ADAMTS-4, and all of them were secretory proteins and could be distributed in the cerebral cortex. ADAMTS-2, ADAMTS-3, ADAMTS-10, ADAMTS-16 contain the functional domains of ADAMTS-4, and the secondary structure within the functional domain is similar. [Conclusion] The proteins ADAMTS-2, ADAMTS-3, ADAMTS-10 and ADAMTS-16, which may have the effect of shearing the Reelin, were screened out.
