目的:用超高效液相色谱法同时测定钩藤中的钩藤碱和异钩藤碱的含量。方法:以ACQUITYUPLCR BEH130C18(2.1mm×100mm,1.7μm)为色谱柱,乙腈-0.01mol/L三乙胺水溶液(45:55,冰醋酸调pH7.0)为流动相,检测波长为245nm,流速0.2mL/min。结果:钩藤碱在0.052-0.309μg具有良好的线性关系,r=0.9997,平均回收率为98.3%,RSD为0.88%,异钩藤碱在0.060μg-0.363μg具有良好的线性关系,r=0.9999,平均回收率为97.7%,RSD为1.29%。结论:该方法简便快捷、精密度高、结果准确。可用于钩藤的质量控制。
OBJECTIVE: To simultaneously determine the contents of rhynchophylline and isorhamnetine in Uncaria rhynchophylla by ultra performance liquid chromatography. METHODS: The mobile phase consisted of ACQUITY UPLCR BEH130C18 (2.1 mm × 100 mm, 1.7 μm) column, acetonitrile-0.01 mol / L triethylamine aqueous solution (45:55, glacial acetic acid, pH7.0) 0.2 mL / min. Results: Rhynchophylline had a good linear relationship in the range of 0.052-0.309μg, r = 0.9997, the average recovery was 98.3%, the RSD was 0.88%, and the isoineurine in 0.060μg-0.363μg had a good linear relationship, r = 0.9999, the average recovery was 97.7%, RSD was 1.29%. Conclusion: The method is simple, rapid, precise and accurate. Can be used for Uncaria quality control.
