目的寻找适合脑内多种神经活性物质免疫组织化学研究的固定方案。方法以胆碱乙酰化酶、P物质和γ-氨基丁酸为递质合成酶、多肽及氨基酸递质的代表,以大鼠隔-斜带-基底核此三类阳性神经元的数量、染色和形态变化为依据,检测了一系列复合固定配方对此三类抗原免疫组化的影响。结果经0.2%戊二醛、4%多聚甲醛和0.2%苦味酸灌注固定和4%多聚甲醛后固定,并经1%硼氢化钠处理的脑组织,能很好地显示该三类神经元。此结果还得到猫脑免疫电镜研究的支持。结论有可能设计一种满足脑内尽可能多种类递质及合成酶免疫组化光、电镜研究所需的复合固定配方,以期在同一实验动物获得最大程度的化学神经解剖学信息。
Objective To find a suitable protocol for the immunohistochemical study of many neuroactive substances in the brain. Conclusion It is possible to design a composite fixed formulation that meets the needs of many types of neurotransmitters and synthetases in the brain for light and electron microscopy studies in order to obtain the maximum chemical neuroanatomical information in the same experimental animal.
