利用显微注射法制备了人βE-珠蛋白基因的转基因小鼠系,其外源基因(βE)能通过生殖细胞稳定地传递给子代,具有典型转基因鼠特征。证实人βE-珠蛋白基因在转基因鼠中的高水平表达需有红系增强子结构(5′HS2)存在。单一位点整合过多拷贝(16拷贝)人5′HS2βE-基因的平均表达水平明显低于较少拷贝(2拷贝)整合的表达水平。
The transgenic mouse line of human βE-globin gene was prepared by the microinjection method. The foreign gene (βE) can be transmitted to the offspring stably by the germ cells, and has typical characteristics of transgenic mice. Confirming the high level of human βE-globin gene expression in transgenic mice need erythroid enhancer structure (5’HS2) exist. The average expression level of the human 5’HS2βE-gene was significantly lower than that of the less copy (2 copies) of the single-site overcomplexed copy (16 copies).
