目的:观察壮骨止痛方提取物对去势大鼠Dkk1蛋白的影响,并探究其治疗骨质疏松症的具体机制。方法:60只3月龄的健康雌性SD大鼠,随机分为壮骨止痛方组、模型组、假手术组、空白组,每组15只。壮骨止痛方组、模型组去势(去卵巢)后建立绝经后骨质疏松大鼠模型,假手术组去除卵巢周围相应体积脂肪。术后5d开始给药,13周后取血和骨组织,测定血清中雌激素(E2)、降钙素(CT)、骨钙素(BGP)含量变化、骨组织病理变化、骨小梁密度,以及骨组织中Dkk1、β-catenin蛋白的表达情况。结果:与模型组相比,壮骨止痛方能降低去势雌鼠血清BGP、CT含量,差异具有统计学意义(P<0.05);壮骨止痛方组E2与模型组相比,差异具有统计学意义(P<0.05)。壮骨止痛方组相比模型组,骨髓腔扩大不明显,且骨小梁形态较为正常;与模型组相比,壮骨止痛方组骨小梁密度有明显升高,具有显著性(P<0.05);与模型组比较,壮骨止痛方组骨组织中骨组织Dkk1蛋白的表达降低、而β-catenin蛋白的表达增多,差异具有统计学意义(P<0.01)。结论:壮骨止痛方可能是通过下调去势雌鼠体内Dkk1蛋白表达,进一步直接影响β-catenin的表达,间接影响雌激素、降钙素等激素的变化,从而达到改善骨代谢紊乱来治疗骨质疏松症。
OBJECTIVE: To observe the effect of Zhuanggu Zhitong Recipe on Dkk1 protein in ovariectomized rats and to explore its specific mechanism of treatment of osteoporosis. Methods: Sixty healthy three-month-old female Sprague-Dawley rats were randomly divided into two groups: model group, model group, sham operation group and blank group. Zhuangguzhitongfang group, model group, castrated (ovariectomized) rat model of postmenopausal osteoporosis, sham operation group to remove the corresponding volume of fat around the ovary. The rats were sacrificed at 5 days after operation and the blood and bone tissues were taken after 13 weeks. The content of estrogen (E2), calcitonin (CT) and osteocalcin (BGP), the pathological changes of bone tissue, trabecular bone density , As well as the expression of Dkk1 and β-catenin in bone tissue. Results: Compared with the model group, Zhuangguzhitong Decoction can reduce serum BGP and CT in ovariectomized female rats, the difference was statistically significant (P <0.05); Compared with the model group, the difference was statistically significant Significance (P <0.05). Compared with the model group, Zhuangguzhitongfang group did not have obvious expansion of the bone marrow cavity and the trabecular shape was normal. Compared with the model group, the density of trabecular bone was significantly increased in the Zhuangguzhitongfang group (P < 0.05). Compared with the model group, the expression of Dkk1 protein in the bone tissue of the Zhuangguzhitong Recipe group was decreased, while the expression of β-catenin protein was increased. The difference was statistically significant (P <0.01). Conclusion: Zhuanggushi Tongfang Prescription can reduce the expression of Dkk1 protein in the female rats and further directly affect the expression of β-catenin, indirectly affect the hormones such as estrogen and calcitonin, so as to improve the bone metabolism disorders to treat the bone Osteoporosis.
