目的探讨结肠癌患者Wnt信号通路的变化情况及其作用意义。方法利用Human Ge-nome U133Plus2.0基因芯片对结肠正常黏膜组织和癌组织基因表达谱进行检测,分析Wnt信号通路基因表达变化情况。结果Wnt信号通路中FRP、Frizzled、CK2、p53、PP2A、CKIα、JNK、CaMKII、CaN、c-jun、cyc-D等关键基因均呈下调,调控着细胞周期、基因转录等。结论Wnt信号通路可能是调控结肠癌演变过程的重要信号通路之一。
Objective To investigate the changes of Wnt signaling pathway in patients with colon cancer and its significance. Methods Human Ge-nome U133Plus2.0 gene chip was used to detect the gene expression profiles of normal mucosa and carcinoma of colon, and the changes of Wnt signaling pathway gene expression were analyzed. Results The key genes such as FRP, Frizzled, CK2, p53, PP2A, CKIα, JNK, CaMKII, CaN, c-jun and cyc-D were all downregulated in Wnt signaling pathway, which regulated cell cycle and gene transcription. Conclusion Wnt signaling pathway may be one of the important signal pathways that regulate the evolution of colon cancer.
