目的探讨白蚁菌圃抗炎的作用机制。方法采用角叉菜胶致正常及切除双侧肾上腺小鼠足肿胀;角叉菜胶致大鼠胸膜炎;蛋清致小鼠足肿胀的炎症模型。分别用酶联免疫吸附法、紫外分光光度法和荧光分光光度法测炎性渗出液中白三烯(LTB4)、前列腺(PGE2)、5-羟色胺(5-HT)及组胺的水平。结果角叉菜胶致正常组及去肾上腺组小鼠足肿胀炎症组织及血清中PGE2含量与阴性对照组比有显著性差异;在蛋清致足肿胀中的组胺和5-羟色胺的含量与阴性对照组比有显著性差异;在角叉菜胶致胸膜炎大鼠胸腔渗出液中白三烯含量与阴性对照组比有显著性差异。结论白蚁菌圃的抗炎作用可能与其抑制炎症部位PGE2、LTB4、5-HT及组胺的水平有关,其抗炎作用不依赖下丘脑-垂体-肾上腺皮质系统的调节。
Objective To investigate the anti-inflammatory mechanism of termite fungi. METHODS: Carrageenan-induced normal foot swelling and paw swelling in bilateral adrenal glands; carrageenan-induced pleurisy in rats; and inflammation in mouse feet caused by foot swelling. The levels of leukotriene (LTB4), prostate (PGE2), 5-hydroxytryptamine (5-HT) and histamine in inflammatory exudate were measured by enzyme-linked immunosorbent assay, ultraviolet spectrophotometry and fluorescence spectrophotometry, respectively. Results There were significant differences in the content of PGE2 in the foot edema and inflammatory tissue and the serum in normal and unadulterated mice compared with the negative control group; the content and the negative correlation between histamine and 5-hydroxytryptamine in the foot swelling caused by egg white. There was a significant difference in the control group; there was a significant difference in the leukotrienes content in the thoracic exudates of carrageenan-induced pleurisy rats compared with the negative control group. Conclusion The anti-inflammation effect of termite bacterial nurseries may be related to the inhibition of inflammation sites PGE2, LTB4, 5-HT and histamine levels, and its anti-inflammatory effect does not depend on the regulation of the hypothalamic-pituitary-adrenocortical system.
