本研究旨在揭示重组人白细胞介素-3对恶性血液病细胞分裂指数的影响及白血病、MDS刺激增生反应率和异常核型出现频率,进而为临床在白血病和MDS化疗造成骨髓抑制时可否应用IL-3提供依据。1 材料与方法1.1 研究对象 1992~1997年确诊的36名患者中,MDS 16例,ANLL 15例,CML 4例,非霍奇金淋巴瘤合并粒-淋白血病1例。男29例,女7例,中位年龄28.2岁(2~70岁)。1.2 方法 条件培养基的制备:将rIL-3(美国Immunen Corporation产品)加入配制的RPMI1640培养基中,终浓度为10U/mL,放置-20C冰箱备用。骨髓细胞培养及染色体的制备:每例分3种培养条件制备染色体。即①条件培养基+5Fdu同步化法:取患者骨髓,按(1~3)×10~6/ml有核细胞数接种于5ml条件培养基中,培养24 h后,常规5-Fdu同步化法制备染色体,GTG显带。②常规5-Fdu同步
This study was to investigate the effect of recombinant human interleukin-3 on the mitosis index of hematological malignancies and the frequency of leukemia, MDS-stimulated proliferation, and the frequency of abnormal karyotypes, which in turn can be used clinically to determine whether myelosuppression caused by leukemia and MDS can cause bone marrow suppression. IL-3 provides the basis. 1 Materials and Methods 1.1 Subjects Of the 36 patients diagnosed from 1992 to 1997, 16 were MDS, 15 were ANLL, 4 were CML, and 1 was non-Hodgkin’s lymphoma with granulocytic leukemia. There were 29 males and 7 females with a median age of 28.2 years (range, 2 to 70 years). 1.2 Methods Preparation of conditioned medium: rIL-3 (Immunen Corporation, USA) was added to the prepared RPMI1640 medium to a final concentration of 10 U/mL, and the -20C refrigerator was set aside. Bone marrow cell culture and chromosome preparation: Chromosomes were prepared for each case in three culture conditions. That is, 1 conditioned medium + 5Fdu synchronization method: take the patient’s bone marrow, inoculate (1 ~ 3) × 10 ~ 6/ml nucleated cells in 5ml conditioned medium, after 24h culture, regular 5-Fdu synchronization Preparation of chromosomes, GTG banding. 2 regular 5-Fdu synchronization
