目的 研究大鼠局灶性脑缺血再灌注期细胞间粘附分子1( I C A M1) 、肿瘤坏死因子α( T N Fα) 在转录水平的表达规律及相关性。方法 用插线法建立大鼠局灶性脑缺血再灌注模型,用逆转录聚合酶链式反应( R T P C R) 测定缺血再灌注期 I C A M1m R N A、 T N Fαm R N A 的表达变化。结果 缺血侧皮质 T N Fαm R N A 在缺血后1 小时开始显著升高,至缺血再灌注2 小时达到高峰,持续高表达1 周; I C A M1m R N A 在缺血后2 小时开始显著升高;至缺血再灌注10 小时达到高峰,持续高表达1周。二者具有密切的相关性。结论 I C A M1 、 T N Fα转录水平在脑缺血再灌注早期表达升高, T N Fαm R N A、 I C A M1 m R N A 的表达具有正相关性。
Objective To investigate the expression of intercellular adhesion molecule-1 (Tcf M) and tumor necrosis factor α (T N Fα) at the transcriptional level in focal cerebral ischemia-reperfusion in rats. Methods The model of focal cerebral ischemia-reperfusion was established by the method of patch-line. Reverse transcription-polymerase chain reaction (RT-PCR) was used to determine the I C A M1m R N A , T N Fαm R N A expression changes. Results T N Fαm R N A in ischemic cortex began to increase significantly 1 hour after ischemia and peaked at 2 hours after ischemia-reperfusion, and remained high for 1 week. I C A M 1m R N A 2 hours after the start of significant increase; to 10 hours after ischemia-reperfusion peaked, sustained high expression for 1 week. The two are closely related. Conclusions The transcription level of I C A M1 and T N Fα is positively correlated with the expression of T N Fαm R N A and I C A M1 m R N A in the early stage of cerebral ischemia / reperfusion.
